Thawing Protocol for 0.5 ml straws of frozen equine semen

 Please note: semen was frozen by Jou Veterinary Services

each straw contains approx. 1 billion sperm cells with at

least 65% progressive motility after thawing.

 

Only ONE straw is needed per cycle !

 

Materials needed

            Water bath at 60o Celsius

            Water bath at 37o Celsius

            Thermometer

            Pair of scissors

            Forceps

            Sterile sleeves and lube, towels, tail wrap

            Bucket of water and disinfectant

            Insemination gun

 

Thawing Procedure

Place the two water baths beside the liquid nitrogen container

Make sure the temperature of the water baths are at 60oC and 37oC respectively

Take one straw out of the dry shipper and immediate immerse the straw into the 60oC water bath

for six (6) seconds by the stopwatch. Hold the straw gently by the tip end with hemostats during this process.

After six seconds, immediate transfer the straw to the 37oC water bath for an additional  fifteen (15) seconds

Remove straw from 37oC water bath after 15 seconds

Remove all residual water from the outside of the straw with a dry paper towel

Remove (cut) the sealed end opposite to the plug

Load the insemination gun

Inseminate the mare (uterine body) immediately

 

Suggested breeding regimen

Being palpating on the second day of heat

Adminster 2500 IU of hCG IV, if possible in the early AM, when the follicle is >= 3.5 cm

(Palpate every 6 hours beginning 24 hours after administering hCG – please note, we do not  palpate

every 6 hours  here at the farm, we just inseminate 36 hours after administering hCG)

The mare will probably be ready to breed the following evening, i.e. 36 hours after administering

hCG. 85% of mares will ovulate within 36 – 48 hours after administering hCG on this size follicle.

When ovulation is imminent, inseminate using one straw of semen, depositing it slowly into the body of the uterus.
